Media - YEM (YEast extract - Manitol)

This is a very basic recipe used to grow rhizobium strains.

Composition:

1 liter 2 liter 4 liter 5 liter
Mannitol 5 g 10 g 20 g 25 g
Yeast extract 0.5 g 1 g 2 g 2.5 g
MgSO4 x 7 H20 0.2 g 0.4 g 0.8 g 1 g
NaCl 0.1 g 0.2 g 0.4 g 0.5 g
K2HPO4 0.5 g 1 g 2 g 2.5 g
Na Gluconate 5 g 10 g 20 g 25 g
add water to 1 l 2 l 4 l 5 l


Prepare 16.6% CaCl2 solution.

Autoclave for 20 min at 120 °C

Add 1 ml of the 16.6% CaCl2 solution to 1 liter YEM medium (1:1000)

Let cool to ca. 55 °C before adding antibiotics

Store at room temperature or 4 °C

Plates:

Prepare YEM as described above, but add before autoclaving 1,5 % Agar (15 g per liter)

Materials needed:

Yeast Extract (# Y1333.1000) by Duchefa
