Media - YEM (YEast extract - Manitol)

This is a very basic recipe used to grow rhizobium strains.
Composition:
1 liter | 2 liter | 4 liter | 5 liter | |
Mannitol | 5 g | 10 g | 20 g | 25 g |
Yeast extract | 0.5 g | 1 g | 2 g | 2.5 g |
MgSO4 x 7 H20 | 0.2 g | 0.4 g | 0.8 g | 1 g |
NaCl | 0.1 g | 0.2 g | 0.4 g | 0.5 g |
K2HPO4 | 0.5 g | 1 g | 2 g | 2.5 g |
Na Gluconate | 5 g | 10 g | 20 g | 25 g |
add water to | 1 l | 2 l | 4 l | 5 l |
Prepare 16.6% CaCl2 solution.
Autoclave for 20 min at 120 °C
Add 1 ml of the 16.6% CaCl2 solution to 1 liter YEM medium (1:1000)
Let cool to ca. 55 °C before adding antibiotics
Store at room temperature or 4 °C
Plates:
Prepare YEM as described above, but add before autoclaving 1,5 % Agar (15 g per liter)
